چکیده

Two distinct tRNA (m G) methylases have been found in the yeast Saccharomyces cerevisiae. They differ in their chromatographic properties on hydroxyapatite, in their response to spermine, and in their site specificity. Only one of the methylases is active against normal tRNA from Escherichia col

Detection of yeast ribosomal RNA sequences in E. coli infected with hybrid bacteriophage.

Yeast ribosomal DNA was inserted into Escherichia coli on a bacteriophage vector and the host cell RNA was then extracted and analyzed for the presence of yeast ribosomal RNA sequences. RNA complementary to yeast rDNA was detected by hybridization. The transcription of yeast rDNA was found to be independent of phage RNA synthesis and to occur on the same DNA strand as rRNA transcription in yeas...

Site-specific AT-cluster insertions in the mitochondrial 15S rRNA gene of the yeast S. cerevisiae.

By comparing the mitochondrial 15S rRNA sequences of four wildtype yeast strains together with their respective secondary structures, with those of the 16S-like ribosomal RNA from other organisms we detected two optional and two invariant AT-clusters. The origin of these clusters is discussed with respect to their roles as possible mobile elements.

Assignment of a yeast protein necessary for mitochondrial transcription initiation.

Yeast mitochondrial DNA contains multiple promoters that are responsible for expression of its genes. The basic yeast mitochondrial promoter consists of a nonanucleotide consensus sequence [5'-ATATAAGTA(+1)-3'] that must be recognized by transcription proteins, including mitochondrial RNA polymerase and any relevant trans-acting factors. Since mitochondrial RNA polymerase alone appeared unable ...

Site specific enzymatic cleavage of RNA

The hybridization of a DNA oligonucleotide a specific tetramer or longer) will direct a cleavage by RNase H (EC 3.1.4.34) to a specific site in RNA. The resulting fragments can then be labeled at their 5' or 3' ends, purified, and sequenced directly. This procedure is demonstrated with two RNA molecules of known sequence: 5.8S rRNA from yeast (158 nucleotides) and satellite tobacco necrosis vir...

Transcription and RNA processing of mammalian genes in Saccharomyces cerevisiae.

The recognition of mammalian genes encoded within a mouse yeast artificial chromosome (YAC) by the yeast transcription and RNA processing machinery was investigated. Transcripts from five genes known to be encoded by the YAC were all found in the total yeast RNA. Of 12 mouse introns assayed, six were correctly spliced by the yeast. However, an abnormal transcription of mouse DNA was also observ...
